    I didn't quite understand the snake since we haven't really seen snakes on the show before. Maybe it has something to do with Sam not having a soul since instinct tells humans to be afraid of snakes ?

    The snake is the archetypal (Biblical) representation of evil, corruption, and sin. See: Adam & Eve.

    The snake is an obvious reference to the corruption of Sam, but it looks lik Dean has a bit of a halo.
